We're pleased to announce that Nimbus has been selected for AWS's elite Building with Llama program, one of 30 companies chosen from over 1,000 applicants! This program provides access to advanced AI models from Meta, running on AWS infrastructure, to accelerate our drug discovery efforts. We’re fine-tuning these tools to enhance our chemistry-aware AI infrastructure to help our scientists explore chemical space more efficiently, including predicting molecular properties, assessing drug-like characteristics, evaluating synthetic routes, and analyzing compound-target interactions. Our thanks to AWS Startups, Meta for Developers, and Azadeh Yazdan for this opportunity, and congratulations to our fellow program participants.

We’re excited to share new research from our team, published in the Journal of Medicinal Chemistry: “Knowledge and Structure-Based Drug Design of 15-PGDH Inhibitors” Our team solved the first cocrystal structure of 15-PGDH bound to small molecule inhibitors, revealing new opportunities for rational drug design. We combined structure-based design with advanced computational methods and machine learning to rapidly identify potent inhibitors. The lead compound demonstrated dose-dependent elevation of PGE2 in vivo—a critical mediator in immune function and tissue regeneration. Congratulations to the authors for this achievement! We’re excited to be at the American Conference on Pharmacometrics (#ACoP2025) in Aurora, Colorado from October 18-21. Our own zhenhong li will be presenting a poster: "Applying Mechanistic PKPD Models to Differentiate WRN Inhibitors Based on Predicted In Vivo Target Engagement." Using computational modeling, the team compared covalent and non-covalent compounds targeting WRN, a synthetic lethal target in MSI-H cancers, and shows how optimized non-covalent inhibitors can achieve comparable or better target engagement while offering potential safety advantages. Our team is headed to ESMO - European Society for Medical Oncology in Berlin, Oct 17-21, where we will share the study design for NDI-219216, our investigational non-covalent WRN inhibitor that has been shown to be selective and potent in preclinical studies and is currently recruiting and progressing through Phase 1 clinical development for the treatment of MSI-H solid tumors.
